An eyewitness said a floor gave way during a concrete pour on the mixed-use development in Victoria Street, at about 10.40 this morning.
Main contractor on the job is Turkington, which is partnering with Laing O'Rourke on the Northern Ireland school framework.
Some workers are reported to have fallen 30ft to the ground and become trapped under the debris. The injured people were taken to the Royal Victoria Hospital.
Another eyewitness said injured people were "walking about covered in dust, holding their heads, dazed".
The project involves the demolition and redevelopment of the existing building for client The Law Society, providing a mixture of retail and office units. It is valued at £8.7m.
